摘要:
旨在研究紫云英苷对雷公藤甲素诱发小鼠睾丸损伤的保护作用,通过建立雷公藤甲素诱发小鼠睾丸损伤模型,采用紫云英苷(15、30和60 mg/kg)进行预处理治疗,检测小鼠睾丸指数、组织形态学、睾丸内丙二醛(MDA)水平和超氧化物歧化酶(SOD)活性; 采用原位末端缺刻标记法(TUNEL)检测睾丸组织凋亡情况; 免疫组织化学和免疫印迹法检测睾丸组织磷酸化JNK(p-JNK)的表达。研究表明:紫云英苷可以有效地保护雷公藤甲素所导致的睾丸组织损伤,缓解睾丸内氧化应激状态从而减少睾丸细胞凋亡。本研究结果将为紫云英苷进一步被开发成为治疗男性生殖系统疾病的药物提供理论基础。
Abstract:
In this study,we demonstrated the protective effect of astragalin in triptolide induced testicular damage models.The results of testicular index and histopathological changes of testes illustrated that treatment with astragalin(15, 30 or 60 mg/kg)led to a significant reduction in testicular damage.SOD activities and MDA level measured by chemical colorimetry showed the antioxidant capacity of astragalin.TUNEL assay revealed that astragalin significantly reduced the apoptosis rate in mouse testes.Also,the phosphorylation of JNK was significantly declined in astragalin groups.Our study has identified astragalin as a novel agent that contributes to the protective activity in testicular damage.
