摘要:
通过PCR克隆了一种来自枯草芽孢杆菌str.168菌株的甘露糖-6-磷酸异构酶编码基因(BSMPI-2),利用pET-28a载体转化至大肠杆菌BL21(DE3)菌株。经IPTG诱导实现了BSMPI-2蛋白的表达,经过亲和层析纯化和聚丙烯酰胺凝胶电泳(SDS-PAGE)验证,证实目的蛋白分子量约3.54×104,BSMPI-2以L-核糖为底物时的最适反应条件为pH 8.0、60 ℃、外源添加0.5 mmol/L Co2+,该酶催化2 h可将29%的L-核糖异构为L-核酮糖。
